相比近年来有长足进展的生物样品纯化和晶体X射线衍射技术,生物大分子晶体的培养依然是一个试验探索的过程,极大程度地依赖于经验并带有较大的盲目性。 蛋白结晶所需的各种成分,如沉淀剂、盐、缓冲液体系、有机溶剂以及各种添加剂,构成一个庞大的多维集合。虽然现在市场上有一些商业化的晶体生长条件筛选盒,但它们只能构成一个一个独立的小子集, 远远无法覆盖非常复杂、巨大的蛋白结晶条件集合。
博亚的BioXtal™结晶条件筛选盒是为蛋白晶体生长条件筛选而特别开发的一个晶体生长条件组合。 其成分组成是以大量发表和未发表的结晶统计数据为基础发展而成。它既包含离散矩阵筛选模式也包括系统性筛选模式,既含*效的结晶条件也含许多其它结晶条件筛选盒所未有的条件。 扩展了筛选范围从而增大了发现结晶条件的几率。
该试剂盒有96个条件以适合96孔板自动化筛选设备,pH范围涵盖蛋白晶体最易生长范围4.5-8.5。所使用的化学试剂来自国标*化学试剂公司(Fluka、Sigma-Aldrich等),并且每种溶液都经过0.22μ虑膜过虑灭菌处理。96个条件分别密封在螺旋盖试管中,每管不少于10毫升,可随便使用。
在最近进行的大量蛋白晶体筛选测试研究中,博亚BioXtal™结晶条件筛选盒曾结晶出了超过50%的可结晶蛋白。被认为是高效结晶条件组合,是一个对现有商业化的结晶条件筛选盒的有力补充和发展。
